Not a cartoon, but a webcomic - A Softer World leaves me either laughing or feeling vaguely disturbed on a weekly basis.
It ranges from straight forward funny strips like this one, to what seem like twisted visual haikus like this, this and this.
Speaking strictly of cartoons, newgrounds.com ranges between the completely awesome and the completely awful with some of their original animated shorts.
As for webcomics, Something Positive (www.somethingpositive.net) is one of my favorites, as is Venus Envy (http://venusenvy.keenspace.com) and Scary Go Round (www.scarygoround.com).
So do we mean animation or comic strips? Strangely enough, they're not entirely interchangable...
In toons I have to say Weebl and Bob out of loyalty but I also dig on Homestar Runner.
As regards comic strips I enjoy Maakies, Kaz's Underworld and Red Meat, but all of these occasionally surface in print form, so I'm not sure if they count. There's a big place in my heart for Penny Arcade, but it's lit only by a tv screen, and smells slightly of wet carpets and Lynx Africa.
I read a couple...Goats, Superosity, and PVP are probably my favourites. All three have been running quite a while, and have complete archives, so you've ample material to waste about a week of your life reading.

DieselSweeties is good, though you'll have to read from the first issue to get most of it. In terms of cartoons, there can only be Retarded Animal Babies.
I can't believe nobody's mentioned Sinfest yet. Wapsi Square is also good, as is VG Cats. Cigarro & Cerveja is a well drawn comic of canadian origin, as is Filibuster, which is a good modern take on the classic one-panel political cartoon. Case Yorke's Gluemeat amuses some, annoys others. Home-grown UK talent can be found at Housd (which you'd probably need to read the archives of to really enjoy and understand) and at Able & Baker. Achewood isn't the best drawn comic out there, but it makes me smile. As do Applegeeks and Mac Hall. Bunny is just plain cute, as is Count Your Sheep. Sam & Fuzzy has a pretty unique style and brand of humour, and Her! Girl vs. Pig is homicidal to the max. Jason Pultz's Comic Strip features a scary bear. Keith Knight's K-Chronicles has a good mixture of political satire and good storytelling. Last but not least, Real Life and Commissioned are worthwhile reads as well (Commissioned has only just started, but looks promising).
On a side note, I am greatful for the -not safe for work- warnings, thanks, I have been victom of looking at a bad link with my boss right around the corner, never a good thing.

Does anyone have a working link to the "Bingo the Clowno" CG animated short? I first saw it about a year ago and now I can't find it anywhere. It is among the creepiest, freakiest, most painstakingly well-produced things I've ever seen online.
On a much less funny and more disturbing tip, The Smoking Gun have Target Practice, the Flash film by Jeff Weise, the kid who shot up his school this week.
Ghastly's Ghastly Comic (NOT safe for work) has its moments. Also, though it's not strictly a webcomic, Frazz is pretty delightful. A very zen comic strip, I think.
I'm a big fan of Player versus Player, especially the way it mixes general soap style storylines with bizarre flights of fancy and various geek parodies.
A definite second for The K Chronicles and I have a big soft spot for Get Fuzzy which is the most consistently funny syndicated strip I have ever read bar Calvin and Hobbes. Webcomix wise, I don't have much to add, and will work through your suggestions.
